Bearing in mind we'll be leaving at 2am, so we shouldn't hit traffic, which route will be quickest?

Just switch your sat nav on and let it take you. Drive slowly and the roads will be quiet.

your best off getting through the traffic on the 62 before half 4 mate. just get there a bit earlier and have an extra pint or 2

Shouldn't take much more than 2 Hours at that time of the morning.

I would go the Stockport way (M60 S/E), personally.
